The NPS has all the stuff on permits and has a basic overview of trails

trails.com has a lot of info, but it seems like you have to register, and maybe pay now.

Various blogs and online journals exist. For example,

You need a wilderness permit, and need to rent or buy a bear-proof container. A bus runs daily in summer between the Valley and Tuolumne (and beyond and back). See http://www.yarts.com http://www.yarts.com/

A week long trip might be a little long for first timers. I'd recommend a 3 night 4 day trip (like a friday to monday trip) instead of a week long one. Taking a shorter first trip will get you used to what you need to pack and what you don't need to pack. Do you know where you want to go?
